abstract:
Matrigel®, a gold standard matrix for organoids culture in vitro, is hardly usable with high-throughput manual/automatic dispensing machines, due to its complex rheology and viscosity dependence from temperature and shear rate conditions. Therefore, we developed an open microfluidic circuit designed to generate 25 Matrigel® domes by exploiting the effect of capillary filling. This module can be then assembled to accomplish a platform for high-throughput personalized medicine on patient derived organoids (PDOs). Computational Fluid Dynamics (CFD) modeling contributes to the description of the filling strategy, related to the 3D features on the bottom of the well, which enable fast complete filling.
